How do I choose cell culture media and supplements?

Pick a basal medium suited to your cell line. RPMI 1640 for a lot of suspension and mammalian lines. Ham's F-12K and others for specific cells. Then supplement it. Most media need these things. Fetal bovine serum (typically 10%), a glutamine source like GlutaMAX and often penicillin-streptomycin and HEPES buffering. What is fetal bovine serum (FBS)? Why is it used in cell culture?

FBS is the liquid fraction of clotted bovine fetal blood. It is rich in growth factors, proteins, hormones and nutrients. It’s added to media at around 10%. It supports cell attachment, growth and proliferation. This is why it's the most used cell-culture supplement. Ocon Chemicals can supply to you defined and standard fetal bovine serum. What is Trypsin-EDTA used for? And how do I passage cells?

Trypsin-EDTA is used to detach adherent cells from the culture vessel. This is so they can be split (passaged) into new flasks. The trypsin breaks cell-surface proteins. EDTA chelates the calcium that aids adhesion. The reaction is then stopped with serum-containing medium. How do I count cells and check viability?

Cell viability is usually checked with the trypan blue dye-exclusion test. Live cells exclude the dye and stay clear. Dead cells take it up and turn blue. Counts are then made on a haemocytometer or an automated counter. Luminescent assays like CellTiter-Glo measure viability by ATP content. What's the difference between PBS, DPBS and Tris buffers?

PBS (phosphate-buffered saline) is an isotonic buffer for washing cells and diluting reagents. DPBS (Dulbecco's PBS) without calcium or magnesium is preferred for rinsing before cell dissociation. Tris-based buffers such as Tris-HCl are used in molecular biology Also in electrophoresis and protein work. Ocon Chemicals has everything in stock. How do I prevent contamination in cell culture?

Contamination from bacteria, fungi, yeast and especially mycoplasma is always a real risk. So what are the main defences. Good aseptic technique in a laminar-flow hood, sterile media and reagents and routine mycoplasma checks. Antibiotics like penicillin-streptomycin add protection but don't exactly replace sterile practice. What is osmolality? Why calibrate osmometers with standards?

Osmolality is the concentration of dissolved solutes in a solution. It’s measured in mOsm/kg. It's a critical quality check for cell culture media, buffers and biopharmaceutical products.
